Tianeptine is an atypical antidepressant, originally developed by the French company Servier in the early 1980s. Compared to more common antidepressant agents that are selective serotonin reuptake inhibitors (SSRIs), Tianeptine works as a selective serotonin reuptake enhancer (SSRE). Up until recently, the monoamine hypothesis was the main theoretical framework used to explain depression and anxiety disorders. Specifically, it is known that serotonin, dopamine and norepinephrine are three main neurotransmitters directly involved in the regulation of motivation and mood. For example, deficiencies in serotonin neurotransmitter are associated with depression and anxiety symptoms. Therefore, the monoamine hypothesis suggests that such imbalances and reductions in certain important neurotransmitters can be restored by reversing monoamine deficits. As a result, the SSRIs work by increasing the levels of serotonin neurotransmitter. In contrast, Tianeptine appears to decrease serotonin levels, which made researchers and clinicians revisit the main theory. One of the biggest disadvantages of using traditional SSRIs is their long list of side effects. However, the same cannot be said about Tianeptine, as it has a much better side effect profile. Although, more research is required, Tianeptine may prove to be a safer and more effective pharmacological treatment of depression and anxiety disorders.
Studies have successfully used Tianeptine in the treatment of depression, anxiety, obsessive-compulsive type behaviours, stress-related disorders and even autistic symptoms. In terms of its nootropic effects (i.e., cognitive functions enhancement), users report improvements in memory, learning abilities and thought clarity following the use of Tianeptine. In addition, Tianeptine has been shown to have neuroprotective properties.
The recommended daily dosage of Tianeptine is 12.5-30 mg, taken once per day. You should not exceed the recommended dosage.
Despite three decades of studies and research, Tianeptine’s mechanisms of action are not fully understood. However, its neuroprotective properties are linked with the ability to prevent pathological changes and glutamate toxicity in the amygdala and hippocampus areas of the brain. Some evidence suggests that Tianeptine can stop and reverse neural damage.
When used within the recommended dosage, Tianeptine is considered to be safe. However, some of the reported side effects include: drowsiness, dry mouth, dizziness, diarrhea, nausea, headache, constipation, fatigue, joint pain and palpitations. In addition, Tianeptine should not be used by pregnant or lactating women, anyone with kidney problems and/or taking other similar substances, namely monoamine oxidase inhibitors (MAOIs) and selective serotonin reuptake inhibitors (SSRIs).
